Abstract

Neoplastic transformation results from a series of genetic alterations involving activation of protooncogenes and inactivation of tumor suppressor genes. Activation of ras protooncogenes by point mutation is the most frequent genetic alteration in spontaneous thyroid tumors. Since these lesions are prevalent in benign and malignant thyroid neoplasm, it has been suggested that they may be an early event in the process of cell transformation. Multinodular goiter and, in particular, goiter nodules, have been considered a true thyroid neoplasm and an interesting model to evaluate the ras proto-oncogene alterations in the pathogeneses of thyroid neoplasia. In accordance, we have demonstrated higher expression of H-ras and K-ras proto-oncogene in nodular tissue, suggesting that control of their expression might play a important role on neoplastic transformation in thyroid cells.
